A new extraction approach for oligonucleotide bioanalysis to enhance recovery, reproducibility and LC-MS sensitivity
Oligonucleotide therapeutics represent a diverse and growing class of drugs with the ability to address disease conditions at the level of gene transcription and translation. As their pipeline expands, the need for quantitative bioanalytical testing via LC–MS has also increased and the challenges to obtaining robust, reproducible quantitative results have become a bottleneck. To address this challenge, Waters is introducing new SPE sample preparation kits that demonstrate improved recovery and reproducibility using a standardized detergent-free protocol that requires little to no method development, is automation friendly and that works across a diverse range of oligonucleotide therapeutics. In this webinar the SPE kits will be introduced and quantitative performance data across a diverse range of oligonucleotide modalities will be presented.
What will you learn?
- Gain an understanding of the LC–MS bioanalytical challenges for oligonucleotides
- How to address critical challenges in establishing an oligonucleotide quantitation workflow: from obtaining selectivity in biological matrices and minimizing oligonucleotide loss due to solubility and non-specific adsorption, to achieving high oligonucleotide extraction efficiency, LC-MS sensitivity and robustness
- How implementation of a new SPE kit-based approach, with QC verified, lot-traceable reagents and standardized protocol, reduces sample preparation complexity, minimizes method development, improves repeatability and facilitates high throughput sample processing
