HEK293 host cell DNA residual testing: transitioning from quantitative PCR to Droplet Digital PCR

This application note explores the process of transitioning from quantitative PCR (qPCR) to Droplet Digital PCR (ddPCR) for HEK293 residual DNA quantification. As immortalized cell lines, HEK293 can be utilized to manufacture biotherapeutic products such as recombinant adeno-associated virus and lentivirus. The transition from the traditional method of qPCR for the qunatification of residual HEK293 is investigated and contrasted with ddPCR technology, a method that is HEK specific, extraction free and does not require standard curves.
